Calcium rises evoked by localized photolysis of caged IP3

Single smooth muscle cell

The smooth muscle cell was voltage clamped in whole cell configuration and the membrane potential maintained at -70mV. Calcium was imaged using fluo-3 at a rate of 25 frames per second and released from the internal calcium store by photolyzing caged IP3. Caged IP3 was introduced to the cell via the patch pipette filling solution. Contraction was prevented by the drug Wortmanin.

Uncaging spots

 

Using the  UGA-40 system for a fast movable Spot Illumination a sequence of 5 spots to be illuminated has been programmed. The delay time between the illuminated spots was 1 sec each, the power of the laser (DPSL 355/30) has been reduced to less than 10%, the pulse was 5ms long. The spots to be illuminated were less than 4 µm and are marked with a red cross in the image below. The calcium response may be watched in the movie below.
